Exchange Boulevard is a city route that has its northern origin at the Four Corners, where State Street turns into Exchange at the intersection with Main Street. Upon emerging from under the Inner Loop, Exchange Boulevard enters Corn Hill and continues south along the Genesee River until it reaches Ford Street, near the bridge. Here it becomes Exchange Street in the Plymouth-Exchange neighborhood.

Exchange also overlaps with Route 383.

Other important intersections are Broad Street, Court Street, Fitzhugh Street, and South Plymouth. The Genesee River Trail also runs parallel to Exchange south of Broad Street.
