East River Road is a very long, meandering route whose northern course runs roughly along the eastern banks of the Genesee River. It originates at West Henrietta Road in Brighton near I-390 and travels south through Henrietta and Rush. It enters the Town of Avon (Livingston County) slightly south of Barber Road, where it turns into Rochester Street.

City of Rochester and Brighton

East River Road begins in Brighton, adjacent to the Laboratory for Laser Energetics at UR's South Campus. It briefly heads west and passes through Genesee Valley Park, which is part of the City of Rochester. It emerges back into Brighton for a short distance before entering Henrietta at Crittenden Road. Kendrick Road is the only major intersection. East River also crosses Red Creek.

Henrietta

East River Road continues along the river and passes the Rochester Institute of Technology.

Major intersections are Route 252/Jefferson Road, Bailey Road, Lehigh Station Road, Route 253, and I-90. It enters Rush at the Rush-Henrietta Townline Road.

Rush

By now, East River is primarily a country road, passing mostly scattered houses and farms.

Intersects with Rush Scottsville Road, Rush West Rush Road and Honeoye Falls 6 Road. Enters Avon just south of Barber Road and changes name to Rochester Road.
